An indispensable tool for low-frequency tumor genetic mutation detection——Double-ended UDI UMI Adapter

 

         At present, tumor genetic testing is mainly divided into two categories according to application scenarios:

One is commonly used to screen tumor genotype,stringently required in the tumor treatment; The other is applied to preventive screening, early screening and recurrence monitoring of the tumor. Thanks to the latter’s befitting healthy people as well, its market potential is huge. Currently, early tumor screening can be performed using Liquid Biopsy, which excels in quick sampling and easy application to large-scale, safe and non-invasive. In order to improve the detection sensitivity of low-frequency mutations and lower false positive rates, Vazyme has now launched the double-ended UDI UMI adapter VAHTS Dual UMI UDI Adapters Set1-Set4 for Illumina (Vazyme #N351-N354) to make your test results more reliable!

It can integrate with deep sequencing to effectively mine low-frequency mutations, so as to facilitate the optimization of detection.

Product Performance:

 

1.Detection of low-frequency mutations as low as 0.1%:

The tumor 0.1% ctDNA standards (including 11 known SNV sites) taken as the template, N351-N354 and like products from I* company are employed to construct the libraries, respectively. Following that, Novaseq 6000 for PE150 sequencing is applied, and the results demonstrate that the quality of sequencing data is comparable to like products from I* company. Based on low-frequency mutation analysis using GATK4 and Vardict software, and library construction using ND607 and ND608, the results demonstrates that the performance of detection of 0.1% low-frequency mutations is better than like products from I* company.

2.Reduction of tag jumping and mismatch:

After premixing 4 or 6 libraries , respectively, the corresponding correlation between different P5 Sample Index (abscissa) and different P7 Sample Index (ordinate) is shown in Figure 4. Color blocks can be used to indicate the probability of different Index combinations. The color blocks on the diagonal correspond to the correct Index combination, and the color blocks in the other parts indicate the Index combination that has tag jump and mismatch. The double-ended UDI sequence can filter the sequences that have had tag skipping and mismatches.

The results show that N351-N354 can effectively reduce the tag jumping and mismatch frequency in hybrid capture libraries.

3.Cross-contamination quality control based on sequencing

The corresponding correlation between different single segments (abscissa) and different UDI (ordinate) is shown in Figure 5. Color blocks are used to indicate the probability of a single segment paired with UDI. The color blocks on the diagonal correspond to the correct combination of a single segment and UDI, and the other color blocks correspond to the wrong combination.

N351-N354 have undergone strict quality control based on sequencing at the factory to avoid cross-contamination. When a single fragment is used to construct a library and then sequenced, the probability of cross contamination is less than 0.01%. Compared with single-ended adapters, the problem of tag jumping and mismatch is greatly reduced, and the problem of a very small number of the tag jumping and mismatch can be solved by double-ended UDI.

 

 

Overview of Vazyme Double-ended Index

Vazyme's latest double-ended UDI UMI adapter VAHTS Dual UMI UDI Adapters Set 1- Set 4 for Illumina (Vazyme #N351-N354) contains three components, namely P5 Index, P7 Index and UMI components. On the basis of the common double-ended adapters Vazyme #N321-N322 (shown on the left side of the figure below), it is equipped with UMI components and contains a specially matched UDI, as shown on the right side of the figure below.
