BCA 蛋白浓度测定试剂盒（BCA Protein Quantification Kit），是目前灵敏度最高的蛋白质测定方法之一。在碱性条件下，蛋白质将Cu2+还原为Cu+，Cu+ 与独特的BCA Reagent A（含有BCA）相互作用产生敏感的颜色反应，形成紫色的络合物。该水溶性的复合物在A562 nm 处显示强烈的吸光性，吸光度和蛋白质浓度在广泛范围内有良好的线性关系，根据吸光值可以推算出蛋白浓度。因此，使用酶标仪测定其在A562 nm 处的吸收值，并与标准曲线对比，即可计算待测蛋白的浓度。

常用浓度的去垢剂SDS，Triton X-100，Tween-20等不影响检测结果， 但受螯合剂(EDTA，EGTA)、还原剂（DTT，巯基乙醇）和脂类的影响。实验中，若发现样品稀释液或裂解液本身背景值较高，可使用Bradford 蛋白浓度测定试剂盒。

Q1：测出值超出标曲范围怎么办？

A1：将样本稀释后再测，摸索一下最适的稀释浓度。

Q2：BCA使用注意事项。

A2：(1) 需96孔酶标板，酶标仪一台，测定波长为A562 nm最佳。也可以使用分光光度计测定，但测定时，需根据比色皿的最小检测体积，适当 加大BCA工作液的使用量使不小于最小检测体积，样品和标准品的用量可相应按比例放大也可不变。使用分光光度计测定蛋白浓度时， 每个试剂盒可以测定的样品数量可能会显著减少。 

(2) 长期不用时，Reagent A/B可置于2-8℃保存，如发现细菌污染则应丢弃。Reagent A试剂在低温条件下出现结晶沉淀时，可37℃孵育使 其完全溶解, 不影响使用。 

(3) 如发现样品稀释液或裂解液本身就有较高背景，请使用诺唯赞生产的Bradford蛋白浓度测定试剂盒。 

(4) 进行BSA标准蛋白标准曲线绘制时，使用样品蛋白的溶解液溶解BSA最优。 

(5) BCA试剂对去污剂不敏感，可兼容SDS 3%、Tween-20 5%、EDTA 15 mM、无EGTA、Triton X-100 5%；对还原剂比较敏感，可兼容 DTT 0.5 mM、β-巯基乙醇 0.07%。 

(6) BCA工作液配制过程中吸取Reagent A液和Reagent B液务必更换吸头，工作液新鲜配制后应立即使用，长时间放置会影响检测灵敏度。 

(7) 最好使用塑料比色皿，如使用玻璃比色皿或石英比色皿，使用后立即用少量95%的乙醇清洗。
